Kisspeptin refers to a family of neuropeptides derived from the KISS1 gene, including kisspeptin‑54, ‑14, ‑13, and ‑10—all sharing a common C‑terminal Arg‑Phe‑NH₂ motif and acting as ligands for the G‑protein coupled receptor GPR54 (also known as Kiss1R). Initially identified as a metastasis suppressor, kisspeptin has since been shown to play a critical role in regulating the hypothalamic–pituitary–gonadal (HPG) axis by stimulating GnRH neurons and initiating secretion of LH and FSH.

Chemical Structure of Kisspeptin

Kisspeptin‑10 (Kp‑10) is a 10–amino acid peptide with the sequence Tyr–Asn–Trp–Asn–Ser–Phe–Gly–Leu–Arg–Phe (YNWNSFGLRF), featuring a conserved Arg–Phe–NH₂ motif crucial for receptor interaction. Its molecular formula is C₆₃H₈₃N₁₇O₁₄, and the molecular weight is approximately 1,302.4 Da. It is also known by several synonyms including KISS‑1, metastin, and Kp‑10 peptide.

What Are the Effects of Kisspeptin?

  • GnRH and Gonadotropin Release: Kisspeptin stimulates GnRH secretion, leading to increased LH and FSH levels in both rodent and human models.
  • Puberty Onset Mechanism: The kisspeptin–GPR54 signaling pathway is essential for initiating puberty. Disruption in this pathway causes hypogonadotropic hypogonadism in animal and human models.
  • Metastasis Suppression: Originally identified as metastin, kisspeptin suppresses cancer cell invasion and metastasis in laboratory studies.
  • Sexual Behavior and Desire: Kisspeptin modulates brain regions associated with sexual arousal and attraction in human trials, providing insight into neuroendocrine-behavior links.
  • Cardiometabolic Regulation: Rodent studies suggest that kisspeptin plays a role in energy homeostasis, appetite regulation, body weight maintenance, and cardiovascular signaling.
