FOXO4-DRI (D-Retro-Inverso) is a synthetic, cell-permeable peptide designed to disrupt the interaction between the FOXO4 protein and the tumor suppressor p53. FOXO4-DRI disrupts this interaction, triggering apoptosis in senescent cells that contribute to aging and a range of age-related diseases. This mechanism positions FOXO4-DRI as a potential therapeutic agent in the field of regenerative medicine and anti-aging research.

Chemical Structure of FOXO4-DRI (15mg)

  • The “DRI” in its name refers to D-Retro-Inverso peptide design: the peptide is composed of D-amino acids in reversed sequence order compared to the original L-amino acid peptide, which increases proteolytic stability and often improves pharmacokinetic properties.
  • It comprises a FOXO4-derived region corresponding to its p53-interaction domain, coupled with cell-penetrating features. In binding studies, it mimics the binding site of FOXO4 (the forkhead domain) to p53’s Transactivation Domain (TAD), especially TAD2.
  • Recent NMR structural studies show that FOXO4-DRI is intrinsically disordered in solution but upon binding p53’s TAD2 region, a transiently folded complex is formed. This involves both hydrophobic and electrostatic interactions.
