What is Hexarelin (5mg)?

What is Hexarelin (Examorelin)?

Hexarelin (also known as Examorelin or EP‑23905) is a synthetic hexapeptide and a potent agonist of the growth hormone secretagogue receptor (GHS‑R1a). It was developed from GHRP‑6 by the addition of two methyl groups to enhance stability and receptor affinity. In research, Hexarelin is studied for its ability to stimulate GH secretion and its metabolic, endocrine, and cardiovascular properties.

Chemical Structure of Hexarelin

Hexarelin’s sequence is His‑D‑Trp(2‑Me)‑Ala‑Trp‑D‑Phe‑Lys‑NH₂, with a molecular formula of C₄₇H₅₈N₁₂O₆ and molecular weight ~887 Da. The peptide incorporates D‑amino acids and a 2‑methyl‑tryptophan residue, which increases stability, bioavailability, and binding affinity to the GHS‑R1a receptor. C-terminal amidation further enhances its biological activity.

What Are the Effects of Hexarelin?

  • Growth Hormone Release: Hexarelin strongly stimulates GH secretion via GHS‑R1a in both human and animal models in a dose-dependent manner.
  • Pituitary and Adrenal Activation: It triggers the release of ACTH and cortisol—likely through arginine vasopressin pathways—and increases prolactin levels.
  • Cardiovascular Effects: Hexarelin enhances left ventricular ejection fraction and binds to cardiac CD36 receptors, showing cardioprotective effects independent of GH stimulation.
  • Appetite and Metabolism: In rats, Hexarelin increases food intake across age groups and supports mitochondrial function during metabolic stress.
  • Aging and Nutritional Sensitivity: GH response to Hexarelin is higher in younger or undernourished subjects and may decrease with age or adequate nutrition.
